Elephant Semen Cryopreservation – Standardization of Techniques for Assessing Sperm Structural Integrity

Development of artificial insemination and semen preservation for elephants requires an improved understanding of sperm biology. Therefore, it is important to develop reliable methods for assessing sperm structural integrity as a means of determining fertilizing ability. This project identified various staining techniques for evaluating plasma and acrosomal membrane integrity, and mitochondrial functionality. It also established an accurate, repeatable and easy to use method for assessment of elephant sperm structural integrity which will aid in the development of elephant semen freezing protocols.
